Nitric oxide (NO) is an extremely important and versatile messenger in biological systems. It has been identified as a cytotoxic factor in the immune system, presenting anti- or pro-inflammatory properties under different circumstances. In murine monocytes and macrophages, stimuli by cytokines or lipopolysaccharide (LPS) are necessary for inducing the immunologic isoform of the enzyme responsible for the high-output production of NO, nitric oxide synthase (iNOS). With respect to human cells, however, LPS seems not to stimulate NO production in the same way. Associação de imunoestimulante com anti-helmíntico no tratamento da verminose em ovinos Ciência Rural

Este estudo teve como objetivo verificar a eficiência do uso de imunoestimulante associado a anti-helmíntico no tratamento das helmintoses de ovinos. Os animais do grupo I (n=29) receberam o anti-helmíntico albendazole (11mg kg-1) em administração única e o imunoestimulante composto de Propionibacterium granulosum (16ug kg-1) e lipopolissacarídeo (LPS) de Escherichia coli (1,2ug kg-1) em duas doses com intervalo de 48 horas e os animais do grupo II (n=29) receberam o anti-helmíntico albendazole (11mg kg-1). Dimethyl sulfoxide effects on the febrile response to lipopolysaccharide injection in rabbits Arq. Bras. Med. Vet. Zootec.

The present study was designed to determine whether DMSO causes an inhibition on the development of fever in rabbits. The intravenous administration of LPS (1.5µg.kg-1 body weight) caused fever in both saline+LPS and DMSO+LPS group, but the onset and magnitude of the induced fever were significantly different. The saline+LPS group presented a prototypic biphasic fever whereas the DMSO+LPS group presented an attenuated febrile response, but it was not abolished. These results suggest that DMSO may provide a protective mechanism against pyrogen LPS, probably through the modulation of NF-kB mediated events, such as fever.

Chronic systemic inflammation and repetitive damage of vascular endothelia by incompatible dialysis system are probable causes of cardiovascular disease in patients on dialysis. The present study aimed to assess in vitro biocompatibility and anti-inflammatory effect of hemodialysis fluid supplemented with rosmarinic acid (RA) using human umbilical vein endothelial cells (HUVEC). Effect of different methods of sterilization on the inactivation of bacterial endotoxin (LPS) in endodontic files BJM

This study evaluated the presence of lipopolysaccharide (LPS) in endodontic files after in vitro contamination with E. coli LPS and sterilization in dry heat or wet heat, with or without previous immersion in a Ca(OH)2 suspension. LPS was quantified using the Kinetic QCL TM test and data were analyzed statistically by the t-test. LPS was quantified only in the contaminated group, not submitted to any immersion or sterilization procedure (0.262±0.1296 endotoxin units/mL) (p=0.0003). In conclusion, both wet heat and dry heat sterilization were effective in inactivating LPS without the need of previous immersion of the files in a Ca(OH)2 suspension.

The esophagus and mouth tumors are very frequent malignancies worldwide. Lipopolysaccharides (LPS) are capable of regulating gene expression of pro-inflammatory cytokines by binding to toll-like receptor 4 (TLR4). Recent studies show that LPS can increase the migration ability of human esophageal cancer cell line HKESC-2 by increasing its adhesion properties. However, the effect of LPS has not been tested on viability of human esophageal and oral cancer cells. Effects of a neuronal nitric oxide synthase inhibitor on lipopolysaccharide-induced fever BJMBR

It has been demonstrated that nitric oxide (NO) has a thermoregulatory action, but very little is known about the mechanisms involved. In the present study we determined the effect of neuronal nitric oxide synthase (nNOS) inhibition on thermoregulation. We used 7-nitroindazole (7-NI, 1, 10 and 30 mg/kg body weight), a selective nNOS inhibitor, injected intraperitoneally into normothermic Wistar rats (200-250 g) and rats with fever induced by lipopolysaccharide (LPS) (100 µg/kg body weight) administration. This study aimed to determine the effects of different concentrations of propofol (2,6-diisopropylphenol) on lipopolysaccharide (LPS)-induced expression and release of high-mobility group box 1 protein (HMGB1) in mouse macrophages. In the present study, histopathological analysis of rat mesentery was used to quantify the effect of two anti-inflammatory agents, dexamethasone (Dex) and pertussis toxin (Ptx), on leukocyte migration. Because low tumor necrosis factor-alpha (TNF-alpha) production has been reported in malnourished children, in contrast with high production of TNF-alpha in experimental protein-energy malnutrition, we reevaluated the production of TNF-alpha in whole blood cultures from children with primary malnutrition free from infection, and in healthy sex- and age-matched controls. Mononuclear cells in blood diluted 1:5 in endotoxin-free medium released TNF-alpha for 24 h. Human T cell lymphotropic Virus type-1 (HTLV-1) induces lymphocyte activation and proliferation, but little is known about the innate immune response due to HTLV-1 infection. We evaluated the percentage of neutrophils that metabolize Nitroblue tetrazolium (NBT) to formazan in HTLV-1 infected subjects and the association between neutrophil activation and IFN-gamma and TNF-alpha levels.
